DESCRIPTION

PEG-MGF (Pegylated Mechano Growth Factor) is a form of IGF-1 (Insulin-like Growth Factor-1) specifically modified through pegylation to extend its half-life and therapeutic activity. Mechano Growth Factor (MGF) is released in response to muscle stress or injury and facilitates muscle repair and regeneration. The pegylation process significantly prolongs the peptide’s presence in the bloodstream, leading to extended anabolic effects and increased muscle recovery and growth.

The product is supplied in the form of a sterile, lyophilized (freeze-dried) powder to be reconstituted with bacteriostatic water for injection.

PHARMACOLOGICAL PROPERTIES

Mechanism of Action: PEG-MGF acts by binding to IGF-1 receptors in muscle tissues, stimulating the activation of satellite cells responsible for muscle repair. This peptide promotes muscle regeneration, reduces muscle breakdown, and enhances muscle growth following injury or exercise. Pegylation helps extend its duration of action compared to unmodified MGF.

Pharmacokinetics:

• Absorption: Following reconstitution and injection, PEG-MGF is absorbed subcutaneously or intramuscularly.
• Distribution: After absorption, it is distributed mainly to muscle tissues where it exerts its effects.
• Metabolism: Pegylation significantly extends the half-life, reducing the rate of breakdown by proteolytic enzymes.
• Excretion: Primarily excreted through the kidneys.

INDICATIONS

PEG-MGF is indicated for:

Muscle Repair and Recovery: Promoting muscle regeneration after injury, surgery, or strenuous physical exertion.

Muscle Hypertrophy: Enhancing muscle growth in athletes or individuals engaged in resistance training.

Treatment of Muscle Wasting Disorders: Supporting muscle maintenance in conditions leading to muscle atrophy (cachexia, sarcopenia, etc.).

DOSAGE AND ADMINISTRATION

Dosage:

• Standard Dosage: Inject 200 mcg to 500 mcg subcutaneously or intramuscularly, 2 to 3 times weekly. The dosage can vary based on individual response, with higher doses administered for muscle repair or hypertrophy.
• For Athletes: Common doses range from 200 mcg to 1 mg per week.

Administration:

Reconstitute the lyophilized powder using the bacteriostatic water provided. Slowly inject the water into the vial and swirl gently until fully dissolved.

Administer the prepared solution subcutaneously or intramuscularly.

Rotate injection sites to avoid localized irritation.

Reconstitution:

• Each 5 mg vial should be reconstituted with 2 ml of bacteriostatic water (supplied).
• Use within 7 days after reconstitution. Store the reconstituted solution in the refrigerator.

   

CONTRAINDICATIONS

Hypersensitivity: Contraindicated in patients with known hypersensitivity to PEG-MGF or any of the formulation components.

Cancer History: Not recommended for patients with active cancer or a history of cancer, as growth factors like MGF may stimulate tumor growth.

Pregnancy and Lactation: Contraindicated in pregnant or breastfeeding women due to potential risks to fetal development.

WARNINGS AND PRECAUTIONS

1. Tumor Growth Risk: Due to its growth-promoting activity, PEG-MGF may enhance the growth of cancer cells. It should be avoided in individuals with malignancies or those at high risk for cancer.

2. Cardiovascular Effects: Careful monitoring is required in individuals with pre-existing cardiovascular conditions due to the risk of fluid retention and increased blood pressure.

3. Hypersensitivity Reactions: Monitor for signs of allergic reactions. Discontinue use if serious reactions such as anaphylaxis or severe skin reactions occur.

4. Renal and Hepatic Impairment: Patients with kidney or liver dysfunction may require dose adjustments due to changes in metabolism or excretion.

ADVERSE REACTIONS

Common side effects:

• Localized reactions at the injection site (pain, redness, swelling)
• Mild fever, fatigue, or flu-like symptoms
• Temporary water retention or edema

   

Serious adverse effects:

• Hypersensitivity: Severe allergic reactions, including anaphylaxis.
• Cardiovascular strain: Hypertension, increased heart rate, or fluid retention.
• Tumor Growth: In patients with underlying malignancies.

INTERACTIONS

Insulin and IGF-1 related treatments: Use with caution in patients on insulin or IGF therapies due to potential synergistic effects on glucose metabolism.

Anabolic Steroids: Co-administration with anabolic steroids may increase muscle mass but also elevate the risk of adverse effects, including cardiovascular strain.

USE IN SPECIFIC POPULATIONS

Pregnancy and Lactation: PEG-MGF is contraindicated during pregnancy and breastfeeding due to potential risks to the fetus or infant. Classified as Pregnancy Category X.

Pediatric Use: PEG-MGF is not recommended for use in children due to the potential impact on natural growth and development.

Geriatric Use: Elderly patients may experience heightened sensitivity to PEG-MGF, particularly with respect to cardiovascular or renal complications. Dose adjustments may be necessary.

 

OVERDOSAGE

In case of overdosage, symptoms such as excessive fluid retention, rapid weight gain, and cardiovascular strain may occur. Supportive treatment should be administered, and monitoring of vital functions is recommended. There is no specific antidote.

STORAGE

Store the lyophilized powder at controlled room temperature (20°C to 25°C / 68°F to 77°F).

Once reconstituted, store the solution in the refrigerator at 2°C to 8°C (36°F to 46°F) and use within 7 days.

Do not freeze. Keep out of reach of children.
